Notch: evidence for combining therapies to treat breast cancer

The Notch signaling pathway has been shown to play a pivotal role in the etiology of breast cancer, as it is aberrantly activated in many forms of the disease, and sustained signaling through the pathway is required to maintain the transformed phenotype. These discoveries have resulted in significant interest in targeting the pathway in breast cancer treatment. Over the past decade, several Notch pathway inhibitors have entered the initial stages of clinical trials as monotherapies.  However, recent data have suggested that Notch pathway inhibitors are most effective when used in combination with conventional breast cancer therapies. This review will explore the role of Notch signaling in breast cancer and how inhibition of this pathway in concert with conventional therapies is developing as a viable treatment option.
